Determination of iridoids in Vitex agnus-castus L. and Vitex cannabifolia Sieb.

Abstract

Leaves and fruits of Vitex agnus-castus L. and Vitex cannabifolia Sieb are reported to be a source of iridoids exhibit dophaminomimetic activity, but iridoid content in their inflorescences and stems is not discovered enough. Also information about iridoids of these plants growing in Ukraine (temperate climate) has not been reported yet. The aim of this study is qualitative and quantitative determination of iridoids in leaves, stems and inflorescences of V. agnus-castus L. and V. cannabifolia Sieb. growing in Ukraine. Each component was determined by methods of thin-layer chromatography (TLC) and high performance liquid chromatography (HPLC). The total iridoid content expressed as aucubin was estimated by UV-spectrophotometric method after reaction with hydroxylamine and iron (III) chloride. By TLC and HPLC methods eight iridoids were found and among them aucubin and agnuside were identificated. The highest aucubin content (4,31 mg/100 g) was determined using HPLC method in V. agnus-castus inflorescences, the highest agnuside content (12,26 mg/100 g) – in its leaves. It was estimated by spectrophotometric method that V. cannabifolia inflorescences and leaves of both species have the higher total iridoid content (2,58–2,87%), expressed as aucubin. This is the reason we recommend leaves of V. agnus-castus and leaves of V. cannabifolia as a new source of iridoids.
