Determination of the content of the main components of the culture medium during the differentiation phase of cell suspensions of Coffea arabica cv. Catimor 9722

Abstract

Exhaustion of culture medium components and possible influence on the in vitro development of plants has been little studied. The current work was developed in order to know and determine contents of the main components in culture medium in the differentiation stage of cell suspensions of Coffea arabica cv. Catimor 9722, during 56 days of culture. Samples of 30 ml were taken every seven days, even from fresh culture medium than the renovated one. Then, content of sugars, 6-BAP and main ions were determined. During the first 21 days of culture and due to the hydrolysis of the sucrose, the two monosaccharide constituents (glucose and fructose) were present. These were completely exhausted at 56 days of culture. Content of 6-BAP in the culture medium diminished quickly during the first 21 days. Although a concentration of 2.75 mg.l -1 was added every seven days, it was not detected in the samples analyzed after 28 days of culture. Content of different ions was determined. Elements such as magnesium and chloride decreased slightly in the culture medium. Contrary happened to phosphate, potassium, calcium and sulphate. Content of ions such as ammonium and nitrate were analyzed and their relationship with the variations of pH too. During the first 21 days of culture there was a further depletion of nitrogen in ammonia form and a rapid decrease in pH. However, after 21 days of culture the first somatic embryos in globular stage were observed. This moment coincided with the beginning of a gradual increment of pH values and also the beginning of nitrogen depletion in nitrate form.
