Distribution of presynaptic neurons of noradrenergic neurons projecting to the spinal dorsal horn in the locus coeruleus of mice

Abstract

Objective To explore the distribution of presynaptic neurons of noradrenergic neurons in the locus coeruleus (LC) projecting to the spinal dorsal horn (SDH) in the whole brain by using the retrograde transsynaptic rabies virus (RV) system. Methods In the first step, the double helper virus was injected into the LC of TH-Cre mice. Two weeks later, the recombinant RV was injected into SDH. After one week of survival, the mice were sacrificed, and then the distribution of upstream nuclei of LC-SDH pathway in the whole brain was observed under laser confocal microscopy. Results By using the retrograde transsynaptic RV system, RV-positive neurons could be observed in the secondary motor cortex (M2), central nucleus of the amygdala ( CeA), lateral periaqueductal gray ( lPAG), ventrolateral periaqueductal gray (vlPAG), dorsal raphe ( DR) and gigantocellular reticular nucleus ( Gi), indicating that these nuclei form a tertiary pathway with LC-SDH pathway. Conclusion The M2, CeA, lPAG, vlPAG, DR and Gi in the brain form a tertiary pathway with LC-SDH pathway.
