Effect Of Different Growth Media, Inkubation Methods And Temperatures On Mycelium Growth Of Shii-Take Mushroom
Journal Title: ISPEC Journal of Agricultural Sciences - Year 2021, Vol 5, Issue 1
Abstract
This study was carried out to determine the effect of different agar media [potato dextrose agar (PDA) and malt extract agar (MEA)] and temperatures (10, 15, 20, 25 ve 30°C) on mycelium growth period and speed of shii-take mushroom. The mushroom spawn and carpophore used in the experiment was provided by Atatürk Horticultural Central Research Institute (Yalova) and Agro Mantar (Denizli), respectively. The prepared MEA (48 g L-1) and PDA (39 g L-1) medium was autoclaved at 121°C for 20 minutes, they were filled into petri dishes under UV light in a sterile cabinet after they became warm and left to cool. After disinfection of stock mushroom carpophores with alcohol, clones were taken from carpophore and one grain was taken from spawn and they were placed on the medium in petri dishes and petri dishes were closed with parafilm. 15 petri dishes from each treatment were prepared and the petri dishes were placed in the incubator at different temperatures. Mycelium lengths were measured regularly from same points and mycelia growth period (day) and speed (mm day-1) was calculated. The main, double and triple interactions of the treatments affected the mycelia development in statistical significance. Among the tested growth media, inoculation methods and temperatures, MEA, grain spawn and 25 °C provided faster mycelia development in a shorter time comparted to other treatments. According to the results of research, 25 °C x MEA x grain mycelia inoculation is found appropriate in the production of shii-take mushroom mycelium.
