Effects of different drying methods on the activity of total flavonoids of Anoectochilus roxburghii(wall.) in vitro

Abstract
"The present paper studied the effects of two drying technologies,vacuum freeze-drying and airdrying, on the antioxidant capacity,tyrosinase activity and melanin biosynthesis of Anoectochilus roxburghii in vitro. The results showed that the content of flavonoids was significantly different in A. roxburghii with two drying methods. The amount of flavonoids was as high as 24.55( ±0.28) mg/g in the vacuum freeze-drying method. In vitro experiments also showed that the antioxidation activity of flavonoids of A. roxburghii within the concentration range of 0.2-5.0 mg/mL was proportional to the flavonoids concentration using vacuum freeze-drying. However,the activity of flavonoids reduced significantly as affected by air-drying method. The antioxidant capacity of flavonoids in freeze-drying Anoectochilus roxburghii( FDAF) was significantly higher than that of the air-drying( ADAF) at the concentration of 5.0 mg/mL with a DPPH clearance rate of 60.16( ±0.005)%,a reducing power of 0.73( ± 0.015),and a total antioxidant capacity of 1.21( ±0.006) mmol Fe2+/mL. The results of human melanoma A375 cells showed that the inhibition rate of tyrosinase activity by FDAF was significantly higher than that of ADAF,but the inhibitory effect on melanin production was significantly lower than that of ADAF,which may be related to the different flavonoid monomers. The experimental results demonstrated that the inhibition rate of tyrosinase activity by FDAF was significantly higher than that of ADAF,but the inhibition of melanin synthesis was significantly lower than that of ADAF in the human melanona A375 cell,which might result from difference of flavonoid monomers. These results provided theoretical basis for Anoectochilus Roxburghii as a valuable ingredient for the foods,cosmetics and pharmaceutical industries."
