Evaluation of genetic diversity within and between populations for Agropyron Trichophorum by ISSR markers

Abstract

This study was carried out using 13 ISSR primers, to genetic diversity evaluation for 17 accessions of 5 Agropyron populations. In total 86 bands (loci) were obtained out of which 84 bands were polymorph in accessions studied. Primer UBC811 with 11 bands had the highest and primer UBC842 with 2 bands had the lowest number of bands. The lowest percent of polymorphism belonged to UBC840 and UBC844 and the percent of polymorphism was 100% for other primers. Average PIC in the used primers was 0.36 that the highest amount of PIC related to primers UBC842 and UBC808 that amount of PIC was 0.47 and 0.45 in this primers and primers UBC811 and UBC823 with the lowest value of PIC don’t have ability in the separation of accessions. Average of MI in the used primers was 2.36 that primers Is11 and Is14 had the highest and UBC842, UBC811 primers had the lowest MI. Semirom population with the Shannon's information index (I) and Nei's gene diversity (He) had the highest variety between reviewed population, and Eghlid population had the lowest variety. The most similarity between populations related to Yasouj and Esfahan (0.87). Cluster analysis for reviewing accession, classified them in 4 groups. Cluster analysis and Scatter plot based on first and second axis from principal coordinate analysis for accessions, showed that genetic variation did not agreement with the geographical distribution.
