Formulation and evaluation of vanishing cream for scalp psoriasis

Abstract
In the present work, vanishing cream based drug formulation was designed and prepared for the treatment of scalp psoriasis. Clinically, psoriasis most frequently affects the skin of the elbows, knees, scalp, lumbosacral areas. It is a typical lesion well-demarcated, pink to salmon-colored plaque covered by loosely adherent scales that are characteristically silver white in color. Keratolytic agents are helpful in reducing scale and hyperkeratosis by causing softening and desquamation of stratum corneum and most beneficial in extremely thick or scaly psoriatic plaques. In the present study salicylic acid chosen as a model drug is the most effective keratolytic agent. These vanishing creams are o/w emulsion based formulation containing suitable combination of oil phase and aqueous phase along with preservatives, prepared and subjected to various physiochemical parameters such as spreadability, tube extrudability, pH, drug content, viscosity and IR studies. In-vitro drug release was carried out in phosphate buffer (pH 7.4) and compared with marketed formulation. Stability studies of a selected formulation were also done at ambient temperature (30oC) for the period of six months as per ICH guidelines. The selected formulations were subjected for primary skin irritation test in rabbits, guinea pigs, and healthy human volunteers for 72 hours and observed for any skin rashes, inflammation, itching, or redness on applied portions. Drug content, pH, spreadability, tube extrudability of the formulation were found to be 98.23%, 6.5, 13.87gm.cm/sec and 97.16% respectively. From rheogram it is concluded that formulation shows pseudoplastic flow property. From our study it reveals that salicylic acid vanishing cream formulations will be useful for the treatment of skin psoriasis. In contrast with ointments which are greasy and messy in nature and may cause staining of cloths. The prepared vanishing cream was pleasant, easily washable thereby increasing patient compliance.
