Hepatoprotective Effects of Extracts of Syzygium cumini Seeds and Citrus sinensis Peels Against Microcystin LR-Induced Toxicity in Rat

Abstract

This study presents the effects of microcystin LR on the liver of rat. Also, the protective effects of jamun seed extract (JSSE) and orange peel extract (OPE) were evaluated on microcystin LR induced hepatotoxicity. Rats were given either microcystin LR (10 μg/kg body wt), microcystin LR (10 μg/kg body wt)+jamun seed extract (200 mg/kg body wt), microcystin LR (10 μg/kg body wt)+orange peel extract (200 mg/kg body wt), orange peel extract (200 mg/kg bod y wt) or jamun seed extract (200 mg/kg body wt) for 30 days. The rats were sacrificed from each group on 15th day and 30th day following the initiation of the experiment. Liver was fixed from each group for light and electron microscopic studies. After 15 day MCLR (group B), MCLR+JSE (group C) and MCLR+OPE (group D) treatment, hepatocytes exhibit hypertrophy and hyperchromatic and hypertrophied nuclei. The sinusoids are dilated. Several shrinked nuclei with irregular boundaries have been noticed indicating degeneration in MCLR treated rats. Degenerated cells are not noticed in MCLR+JSE and MCLR+OPE treated rats. In OPE (group E) and JSE (group F) treated rats no morphological alterations are noticed in hepatocytes. In 30 day MCLR treated (group B) rats, the cytoplasm of hepatocytes become vacuolar. Glycogen deposits are noticed in few cells as well as in sinusoids indicating obliteration in these spaces. Hepatocytes are seen with irregular boundaries with shrunken nuclei. Focal necrosis is recorded at many places. Infiltration of inflammatory cells are seen among necrotic cells. Binucleated cells are also seen. In MCLR treated rats whirling of RER around mitochondria and swelling of mitochondria are visible. In MCLR+JSE (group C) and MCLR+OPE (group D) treated rats the hepatocytes show similar changes as noticed in MCLR treated rats except that focal necrosis and obliteration of sinusoids are not encountered. Also in these rats degeneration of hepatocytes are less as compared to MCLR treated rats. In OPE (group E) and JSE (group F) treated rats the hepatocytes are similar to control and have not shown any noticeable alterations in morphological structure. It can be suggested that microcystins can provoke environmental stress and may pose a potential health ha zard in aquatic organisms specially fish and other vertebrates including human via drinking water and/or food chains.
