HPTLC INVESTIGATION OF PHENOLIC COMPOUNDS FROM THE EXTRACTS OF ULMUS GLABRA HUDS. BARK (ELM)

Abstract

This study was aimed to investigate the crude extracts obtained from bark of Ulmus glabra Huds. (Ulmaceae), in order to identify and quantify some polyphenols using instrumental methods [UV-VIS spectrometry, high performance thin layer chromatography (HPTLC) and densitometry]. We applied different extraction techniques such as: conventional techniques - extraction at room temperature, heat reflux extraction and unconventional technique - ultrasound assisted extraction, varying the main extraction parameters (solid to liquid ratio and time). The resulted crude extracts were quantitatively analyzed by UV-VIS spectrometry and compared taking into account the total polyphenols content (as gallic acid - %mg/mL) and the total polyphenolcarboxylic content (as chlorogenic acid - %g/mL). The quantitative analysis revealed the extracts from bark of U. glabra contain polyphenols (as gallic acid - between 104.76 and 423.30 %mg/mL and as chlorogenic acid - between 0.0503 and 0.2016 %g/mL). The best extracts, obtained using each extraction method, were selected to be analyzed using HPTLC. The qualitative analysis on bark extracts revealed the presence of valuable compounds. HPTLC fingerprinting of extracts showed several peaks, with different Rf values, corresponding to phytocompounds such as chlorogenic acid (Rf = 0.53) and astragalin (Rf = 0.74).The densitometry analysis offers information about the amount of chlorogenic acid (6.84-11.49 % mg/mL) and astragalin (5.36-6.89 % mg/mL). The extracts with the higher content in polyphenols was obtained using heat reflux extraction, a 1:10 solid to liquid ratio and 30 minutes as extraction time. The content in polyphenols suggests the possibilities to use the bark of this species to separate phytocompounds with antioxidant, cytotoxic and antitumoral potential.
