IMMUNOHISTOCHEMICAL STUDY OF E-CADHERIN AND P53 PROTEIN EXPRESSION IN FEMALE MAMMARY CARCINOMAS USING A TISSUE MICRO ARRAY TECHNIQUE GEZIRA-CENTRAL SUDAN

Abstract

Objective: To improve the breast cancer diagnostic and prognostic capabilities by application of immunohistochemical technique for assessment of expression of p53 and E-cadherin, To assess the expression of E-cadherin and p53 protein markers in breast invasive ductal and lobular carcinomas in FFPE (formalin fixed paraffin embedded) tissue. Material and Methods: The material of this study consisted of 41 FFPE blocks of primary breast carcinoma specimens. Multiple sections from all paraffin blocks were cut at 4 microns thickness and stained: 1.Sections from tumors and surrounding breast tissue were stained by hematoxylin and eosin stain for histopathological evaluation. 2.Immunohistochemical staining by E-cadherin monoclonal antibody. 3.Immunohistochemical staining by p53 monoclonal antibody. Results: Out of the 41 cases examined 26cases (63.4%), were positive, while 13 cases (36.6%), were negative, studied 5 cases (12.2%), were negative while 36 cases (87.8%) were positive. Conclusion: Immunohistochemical detection of p53 and E-cad has been demonstrated to be a reliable and easily performed on paraffin embedded tumor specimens, p53 immunostaining is a reliable method for detection of aggressiveness of ductal breast carcinomas and could be considered to have a role in metastatic process in breast carcinoma, p53 protein has a role in E-cad expression in breast carcinomas.
