In-vitro enzyme inhibitory effects of sphaeranthus indicus flower heads ethanolic extract on α-amylase and α-glucosidase enzymes in relation to In-vivo antidiabetic effect on streptozotocin induced diabetic rats

Abstract

The flower heads and leaves of Sphaeranthus indicus are widely used in India and some Asian countries as folk medicine in the treatment and management of a variety of human ailments, including diabetes mellitus. The primary aim of the current study was to study the in vitro inhibitory effects of Sphaeranthus indicus flower heads extracts on the activities of diabetic related carbohydrate metabolizing enzymes α-amylase and α- glucosidase. In addition, the long term (21 days) in vivo effects of Sphaeranthus indicus flower heads ethanolic extrac on fasting blood glucose levelsand other biochemical parameters in normal and Streptozotocin induced diabetic rats were also investigated. Results revealed that Sphaeranthus indicus flower heads ethanolic extract was safe up to 600 mg/kg and dose was selected at 200 mg/kg. Sphaeranthus indicus flower heads ethanolic extract in a dose of 200 mg/kg showed significant anti-α amylase and α-glucosidase activity which is better than other dose. In Streptozotocin induced diabetic rats the ethanolic flower heads extracts of Sphaeranthus indicus showed significant reduction and normalization in increased blood glucose level, AST, ALT, Cholesterol, Triglycerides, LDL, VLDL and elevated the decreased HDL level as that of standard.
