In vitro establishment of Zephyranthes spp.
Journal Title: Biotecnología Vegetal - Year 2017, Vol 17, Issue 1
Abstract
The genus Zephyranthes plants bulbous, herbaceous and perennial, with great ornamental value, because their flowers have a great variety of sizes and colors. The objective of the present work was to in vitro establish of Zephyranthes spp. from seeds and bulbs as initial explants. The seeds were immersed for 10 min in 1.5% sodium hypochlorite. In the bulbs, the outer catheles were removed and cut longitudinally in as many sections (SB) as possible. Three methods of disinfection were evaluated: I. 1 min in ethanol 90% and 20 min in 1.5% sodium hypochlorite. II. 1 min in 90% ethanol and three exposure times, 10, 15 and 20 min in the fungicide Mancozeb (4 g l -1 ). Every seven days the number of explants contaminated with microorganisms was quantified and the percentage of microbial contamination was calculated after 20 days of culture. In the seeds, the number of germinated seeds was quantified and the percentage of physiological germination (protrusion of the primary root) and organ formation were calculated. Bulbs were also evaluated for shoot development up to 20 days of culture. In the seeds microbial contamination was 2% and germination 95%. Organs such as bulbs, roots and leaves were observed. Although shoot were developed, the methods of disinfecting the bulbs were not effective for 100% contamination.

Influence of maltose and sucrose on somatic embryos maturation of Glycine max cv. ‘Incasoy-27’
The somatic embryogenesis in soybean crop (Glycine max) has as limitation the low percentage of conversion to plant that is obtained, attributed to the presence of somatic embryos with abnormal morphologies during the...
Determination of the content of the main components of the culture medium during the differentiation phase of cell suspensions of Coffea arabica cv. Catimor 9722
Exhaustion of culture medium components and possible influence on the in vitro development of plants has been little studied. The current work was developed in order to know and determine contents of the main component...
Influence of growth regulators on callus formation of Phaseolus vulgaris L cv. CIAP 7247
The present work had as objective to achieve the formation of nodular calli in Phaseolus vulgaris L. cv. CIAP 7247 using several growth regulators in the culture medium. Cotyledons of mature seeds without coats were use...
Obtention of microtubers and minitubers as pre basic seed in tree Peruvian potato cultivars
In potato (Solanum tuberosum L.) seed certification programs, the use of in vitro plants, microtubers and minitubers as pre-basic seed are of great importance, especially in countries such as Peru, where there is a nee...
In vitro phosphate solubilization by Aspergillus and Penicillium strains and coffee plants growth promotion (2019)
The availability of phosphorus in the soil for the growth of plants can be increased by the action of phosphate solubilizing microorganisms. The objective of this work was to determine the potential of Aspergillus and...
