In vitro evaluation of the new irrigant cytotoxicity:chlorophyll

Abstract

The disinfection of the root canal system is achieved by the mechanical action of the instruments together with the antimicrobial agents in the irrigating solutions. Many studies are realized with the objective to find an irrigant with antimicrobial activity and tissue-dissolving ability without causing severe inflammatory reactions when placed in contact with vital tissues.The purpose of this study was to estimate the cytotoxicity effects of 100% Chlorophyll by Hep-2 cell (human fibroblast) cultivation.This evaluation was done by cytotoxicity test on cell monolayer by 5 x 105 concentration and exposure time of 5 (five) minutes. Chlorophyll cytotoxic effects were compared with 2,5% sodium hypochlorite used as positive control and Duldecco’s modified Eagle’s medium (DMEM) as negative control.The results were evaluated by Kruskal-Wallis and Mann-Whitney tests, displaying a significant difference (p = 1,2 X 10-10) between them.This study showed us that 100% Chlorophyll solution is not cytotoxic and 2,5% sodium hypochlorite is highly cytotoxic.
