Increased synphilin-1 expression in human elderly brains with substantia nigra Marinesco bodies.
Journal Title: Pharmacological Reports - Year 2008, Vol 60, Issue 6
Abstract
The aim of the present study was to examine the expression of synphilin-1, alpha-synuclein, and tyrosine hydroxylase in human elderly brains and the incidence of Marinesco bodies (MBs, intranuclear inclusions) in the neuromelanin-containing substantia nigra neurons. The brains of twenty-two individuals without clinical signs and symptoms of parkinsonism and dementia and an additional two parkinsonian patients were dissected and subjected to histopathological examination and western blotting. Ubiquitin-positive and agr;-synuclein-negative MBs were found in 0.84-9.45% of the nigral neurons from brains of 15 healthy individuals and both parkinsonian patients. The frequency of pigmented nigral neurons containing MBs was positively correlated with age. The levels of tyrosine hydroxylase in the caudate nucleus and putamen decreased with age, and were inversely correlated with the MB frequency. The level of synphilin-1 in the caudate nucleus was positively correlated both with age and the MBs. Additionally, the MB appearance was correlated with synphilin-1 level in the substantia nigra. No significant correlation between alpha-synuclein expression and age or MBs was found. Our results suggest that synphilin-1 expression increases with aging. Further studies on expression of this protein in elderly brains are warranted.

Brain nitric oxide synthases in the interleukin-1β-induced activation of hypothalamic-pituitary-adrenal axis.
Interleukin-1β (IL-1β), the major cytokine involved in activation of hypothalamic-pituitary-adrenal (HPA) axis modulates both central and peripheral components regulating HPA activity. The role of nitric oxide (NO) gener...
Nitric oxide modulation mediates the protective effect of trazodone in a mouse model of chronic fatigue syndrome.
The present study was conducted with the aim of elucidating the possible role of nitric oxide (NO) in the neuroprotective effects of trazodone used to treat chronic fatigue syndrome (CFS) in mice. Male albino mice were f...
Role of nuclear factor kappaB in the central nervous system.
The aim of this review was to assemble current literature data on nuclear factor kappa B (NF-kappaB). Many authors believe that NF-kappaB, a transcription factor, has essential influence on the regulation of numerous gen...
Accelerated thrombus lysis in the blood of plasminogen activator inhibitor deficient mice is inhibited by PAI-1 with a very long half-life.
Patients with defective plasminogen activator inhibitor protein (PAI-1) or with PAI-1 deficiency can experience hemorrhage as a result of a hyperfibrinolysis. In these patients, a normal thrombus forms, but endogenous ly...
Exposure to alcohol and tobacco smoke causes oxidative stress in rats.
Tobacco smoking and alcohol abuse causes oxidative stress in humans and underlay numerous chronic degenerative diseases. Liver is the main organ exposed to alcohol toxic metabolites, whereas tobacco smoke is chiefly harm...
