Influence of Heat Stress on DNA Damage on Sheep‘S Sertoli Cells

Abstract
It has been proven that the negative consequence which occurs due to heat stress is disruption of DNA structure in male's reproductive tract. So, in order to determine the effect of heat stress on the type of damage caused to the cells under the specified condition, this study has been done to evaluate the size of DNA damage under the heat stress using the TUNEL Sertoli cells at 42 ° C. 9 testes of male lambs ranging 3 to 10 months were taken from the abattoir and the isolation and cell plant of Sertoli cells was carried on these samples,. To count cells which suffered from apoptosis, the TUNEL coloring was performed on positive, negative and control cells. Finally, DNA damage under both mild heat stress and excessive heat stress were measured using Random Design and Statistical One Way ANOVA methods and by the statistical SPSS software. Assessment by TUNEL coloring showed that heat stress has increased the number of damaged cells of DNA and Increase on C º 42 group was statistically significant compared to control group (P<0.05).
