Influence of Metals on Activity of Extracellular Deoxyribonuclease Produced by Aeromonas species

Abstract
Deoxyribonucleases are enzymes that act on deoxyribonucleic acid yielding free nucleotides. Deoxyribonucleases carry out a wide range of reactions including repair, recombination, replication, defense and nutrition. The enzyme industrially is important for genetic engineering applications, antiviral agents, treatment of cystic fibrosis, purification of proteins and many more. Various factors affect the Deoxyribonucleases production which includes pH, temperature, time of incubation and media components. The enzyme requires various metals for the activity. Not a single metal is required for activity by all bacteria rather different metals are required by different types of bacteria. In current study we have characterized an efficient extracellular Deoxyribonucleases producing bacterium from soil on DNA containing media. Further we studied the effect of divalent metal ions on the activity and the species chosen for this purpose was a characterized Aeromonas species. Different concentrations of the metals were used for this purpose. We found that Deoxyribonucleases activity increases when there is presence of manganese & magnesium at 5 mM concentration but continuous addition of metals has shown decrease in the activity of the metals. While cobalt and nickel are the moderate activators of Deoxyribonucleases enzyme produced by Aeromonas species. Calcium and Zinc acts as the inhibitor of Deoxyribonucleases activity of Aeromonas species at all the concentration of metals.
