Investigation and Molecular Pathogen Identification of Sarcocystis from Pigs in Luoyang

Abstract

In order to reveal the infection status and pathogen species of Sarcocystis infection in pork on sale for consumers, 164 fresh pork samples were randomly collected from different supermarkets and vegetable markets in downtown Luoyang. The direct tabletting microscopic examination method was used to observe Sarcocystis cysts firstly. The genomic DNA of cysts positive samples was extracted. Using the genus-specific primers of Sarcocystis based on 18 S rRNA sequence, the genomic DNA of positive samples was amplified by nested polymerase chain reaction ( PCR) . The results show that 44 out of 164 collected samples are positive for Sarcocystis, and the total positive rate is 26. 8%. The degrees of infection are mild or moderate. The cysts in muscle are fusiform or rod-like with a size of ( 367 ~ 1 976) μm × ( 176 ~ 246) μm. All of microscopic positive samples are further identified as Sarcocystis-positive by nested PCR. The PCR products of 21 positive samples are selected for sequencing. The 18 S rRNA sequences of 14 samples are obtained and show the highest similarity with S. miescheriana in the Gen Bank database with 99%. The average positive rate of Sarcocystis in the pork on sale for consumers in downtown Luoyang is high, and the dominant species is S. miescheriana.
