Microbiological justification for the choice of antimicrobic substances concentration in powder composition based on natural zeolite (clinoptyololite)

Abstract

Topicality. Prevention and treatment of skin diseases remain one of the urgent tasks of modern medicine. Powder is one of the effective dosage forms that can be used for this purpose. Powder should be sufficiently adhesive and hold on to the surface of the skin, prevent microbial invasion, has moderate adsorbing properties, reduce the feeling of pain and fail to resolve proteolytic cleavage. At present, there are practically no powder formulations that would meet all of these requirements. Therefore, the issue of creating a new pharmaceutical preparation in the form of powder, having adsorbent, local anesthetic and antimicrobial action is relevant. Aim. To choice the concentration of antimicrobial substances in the composition of powder for external use, by conducting microbiological screening of model samples. Materials and methods. Neomycin sulfate and polymyxin B sulfate were chosen as antimicrobial agents, and natural zeolite (clinoptilolite) was chosen as a filler. For the of antimicrobial activity study, the method of diffusion in agar (method of “wells”) is used. Results and discussion. The experimental data of antimicrobial activity for the model samples of powders indicate the presence of specific antibacterial activity of the samples, and the combination of neomycin sulfate and polymyxin B sulfate in the formulation leads to the potentiation of antimicrobial action. Conclusions. The rational content of neomycin sulfate and polymyxin B sulfate is established at 5000 IU/g and 10 000 IU/g, respectively, and further increase in their concentrations does not lead to antimicrobial activity increase.
