PREPARATION AND CHARACTERIZATION OF DILTIAZEM BASE FROM ITS OFFICIAL SALT FORM

Abstract

 ABSTRACT Diltiazem hydrochloride (DTZ), a drug used in the treatment of angina pectoris is very popular for treatment of old age hypertension. The drug is well absorbed from GI tract, but its bioavailability is low due to extensive first pass metabolism. In general human skin is considered as hydrophobic and the salt form of the original drug has poor permeability as compared to its base form. An attempt has been made to get enhanced release of base form of diltiazem. Base forms of Diltiazem has been prepared and characterized from corresponding salt form. Characterization of pure Diltiazem with EVA(VA 40%) copolymer, Ethyl cellulose,Eudragit RL100 and Eudragit RS 100 was done. FT-IR spectra revealed that there was no Drug-Polymer interactions. DSC thermogram of diltiazem base with optimized EVA co-polymer reveals no interaction. In-vitro drug permeation studies was done for the optimized batch and the permeability was enhanced using azone, propylene glycol and alcohol. The highest flux of diltiazem was found with alcohol.
