NEPHROPROTECTIVE EFFECT OF MELATONIN AGAINST ADRIAMYCIN INDUCED OXIDATIVE RENAL INJURY IN MALE RAT

Abstract

Melatonin, a potent antioxidant molecule, showed to play remarkable role in protecting cells and tissue against oxidative stress. This study investigated the ameliorative effect of melatonin on adriamycin (ADR)-induced kidney injury. Rats were randomly divided into five groups. The first group received no medication and was considered as the control group; the second group was injected ip with a single dose of ADR (10 mg/kg); the third group was treated ip with melatonin (10 mg/kg) for 7 days, the fourth group was treated with melatonin for 7 days before ADR injection and the last group was treated with melatonin for 4 days before and continued for 3 days after the administration of a single dose of ADR. Serum urea, creatinine and uric acid levels were significantly higher after administration of a single dose of ADR than the control group indicating impaired kidney function. The MDA and protein carbonyl levels were significantly higher in the kidneys of ADR-administered group than in the control group. In addition, the activities of SOD, CAT and GSH-Px in kidney homogenates were significantly increased compared to those in the control group. On the other hand, renal GST activity and GSH concentration were significantly decreased in the same animals after ADR treatment indicating renal oxidative stress. Melatonin administration, either before or after ADR treatment, significantly ameliorated the increase in urea, creatinine, and uric acid levels compared to the ADR treated rats indicating improved kidney function. Melatonin also and normalized the activities of SOD, CAT and GSH-Px and reduced MDA and protein carbonyl levels compared with ADR treated group. In addition, the depression in GST activity and GSH concentration in kidney were significantly normalized when MLT was administered either before or after ADR injection. These results depict the effectiveness of melatonin to offer prophylactic role against ADR-induced oxidative stress and reno-toxicity.
