Niesteroidowe leki przeciwzapalne – blaski i cienie

Abstract
Działanie klasycznych niesteroidowych leków przeciwzapalnych polega na hamowaniu syntezy prostaglandyn przez blokowanie obu form enzymu cyklooksygenazy COX-1 i COX-2. Dla milionów ludzi NLPZ odgrywają ważną rolę w leczeniu zespołów bólowych związanych ze stanem zapalnym, są jednak obarczone licznymi objawami niepożądanymi ze strony przewodu pokarmowego, nerek i układu krążenia. Mechanizmy odpowiedzialne za wywołanie powikłań związanych z leczeniem zarówno klasycznymi, jak i selektywnymi NLPZ nie zostały do końca wyjaśnione i nie wydają się polegać tylko na zablokowaniu obu form cyklooksygenazy. W artykule przedstawiono aktualne opinie dotyczące korzyści i ryzyka stosowania NLPZ.
