Photodynamic inactivation of dermatophytes

Abstract

Introduction: Photodynamic therapy is a technique of killing cells using visible light and photosensitizing agent. We demonstrated significant in vitro fungicidal activity against Trichophyton mentagrophytes using laser red light and photosensitisers based on chlorine e6. The aim of study: 1. Photodynamic inactivation of Trichophyton mentagrophytes cells in vitro. 2. Comparison of three varioustypes of photosensitiser (Fotolon, chlorine e6, protoporphyrin IX) for effectiveness PDT method. 3. Comparison of photosensitisers in two various concentration 0.1% and 1%. Materials and methods: The material studied was obtained from patients and placed on mycological bases. Identification was made accordingto standards after the growth strains. Solutions were used immediately after preparation. After preillumination time the following procedures were applied. The laser LS-PDT 662/150 (LaserSecura, Wrocław, Poland) emitting red light at 662 nm wave length and maximum power 150 mW has been used as a light source. Exposure time was 3 minutes 20 seconds, energy density 15 J/cm2. Results: After the first PDT procedure the photodynamic effect was clearly seen in all analyzed cases. After next sessions of PDT the decrease of Trichophyton mentagrophytes was observed. The best result was for samples with protoporphyrin IX. The activity of photosensitisers was similar. Conclusions: This study showed that photodynamic method can be effective to kill T. mentagrophytes cells in vitro. In this study the influence of different concentration of PS's used was no observed.
