PHYTOCHEMICAL EVALUATION OF SCIRPOIDES HOLOSCHOENUS (L.) SOJÁK EXTRACTS

Abstract

Scirpoides holoschoenus (L.) Soják (Cyperaceae) is a little known species, not used in modern therapy, but traditionally used in Romania (in the Danube Delta and the Black Sea) as a decoction to treat digestive, biliary and liver diseases. Different bioactive polyphenols, flavonoids and phytosterols were obtained from the aerial parts and roots of this plant using heat reflux extraction (HRE) and room temperature extraction (RTE). The extracts were analyzed by HPTLC (CAMAG LINOMAT IV, TLC 3 Scanner, WINCATS Planar Chromatography Manager, and stationary phase HPTLC plates G60 F254) and validated as a quantitative method for identifying and quantifying major bioactive compounds with potential uses as antioxidant, anti-inflammatory and hepatoprotective agents. The main parameters for optimal extraction of compounds of interest were: 70%v/v ethanol, heat reflux extraction, extraction condition: solid:liquid ratio = 1/10; 240 minutes, using as raw material the roots and rhizomes. The HPTLC chromatograms revealed a limited range of phenols and phytosterols: chlorogenic acid, ferulic acid, luteolin, β-sitosterol and stigmasterol. Analytical results were obtained for total polyphenols expressed as gallic acid (18.6557-27.0954 % g/g d.w), total flavonoids expressed as rutin (2.3139-2.4290 % g/g d.w.), sitosterols (25.3-49.32 mg/100 mL extract by HPTLC densitometry). These classes of active principles are known for their antioxidant and hepatoprotective activities. The results obtained may serve to promote the use of S. holoschoenus in further research looking at its potential use in biliary and liver disorders in accordance with traditional use references.
