RapidExtract, a novel biodegradable filter cartridge for capture and containment of biomolecules
Journal Title: International Research Journal of Applied and Basic Sciences - Year 2013, Vol 7, Issue 5
Abstract
DNA, RNA and protein isolation is essential in the expansion of science related to the study of these biomolecules. Extraction and purification science has since advanced so rapidly but there seems to be of the same direction in the usage of materials for the capture and containment of biomolecules. There has not been very little attempt to use a biodegradable material to produce the matrix in a column for the capture and containment of biomolecules. In this study, cellulose based biodegradable material was utilized to produce the matrix of an extraction column. Both single-cellular prokaryote and multicellular eukaryote were chosen to be model organisms in the extraction process using a fast extraction solution DeRiProTM . The quality of the extracted biomolecule was tested with spectrophotometry and agarose gel electrophoresis. The isolated product was also used for PCR and restriction enzyme digestion to determine there was no contamination by inhibitors of these processes. The results indicated that the newly developed extraction column using biodegradable material is comparable with three other commercially available extraction kits. Simple and efficient nucleic acid purification by using biodegradable cellulose-based spin column has been designed. This method is able to purified DNA or RNA of different organisms using a single protocol. It is an extraction method with low costing as only two solutions would be used in the whole process. No harmful solvent such as phenol is required and the filtering matrix that used in spin column is biodegradable and renewable.
