Role of PAX9 and MSX1 proteins in dental development and agenesis

Abstract

Introduction: The development of mammalian teeth is a complex process that encompasses a number of epithelial-mesenchymal interactions that involve transcription factors. Due to its complexity, the process is prone to disturbances that may result in tooth agenesis. Transcription factor PAX9 and homeo-domain protein MSX1 are the major of the causative genes in tooth morphogenesis identifiedthus far.Aim of the study: To introduce basic mechanisms of interaction between PAX9, MSX1 and other proteins that are significant for normal or abnormal odontogenesis.Conclusions: Our knowledge of interaction between PAX9 and MSX1 when it comes to regulating gene expression or protein functions is still incomplete. It is possible that PAX9 could interact directly with MSX1 or activate a regulator of MSX1. Alternatively, the molecular relationship shared by PAX9 and MSX1 could involve regulation on the post-transcriptional level. Both proteins may control the expression of BMP4, another gene, whose product is important for proper odontogenesis. Studies of the structure and function of PAX0 and MSX1 genes and proteins are important for advancing our understanding of physiological and abnormal tooth development.
