Screening of metal nanoparticles for application in biotechnology of veterinary immunobiological agents

Abstract

Introduction. Issues specific prevention of animals infections in recent years are particularly important because these diseases are the most common, occurring as a mixed infection, and cause significant economic losses. But given the fact that almost all of these diseases associated with microorganisms, these drugs should be improved with the latest advanced technologies, which include nanotechnology. The goal of the work was to determine biosafe of metal nanoparticles for their using they in the biotechnology of veterinary immunological means against infections of animals. Materials and methods of researches. Metal nanoparticles were synthesized by the F.D.Ovcharenko Institute of Biocolloidal Chemistry, NAS of Ukraine: copper (СuNP) -20 nm; silver (AgNP) - 30 nm; gold (AuNP) - 30 nm; zinc (ZnNP) - 20 nm; iron (FeNP) - 40 nm. Evaluation of cytotoxicity of metal nanoparticles in vitro by the method for assessing cell’s viability with crystal violet and MTT assay was performed using eukaryotic cells line ST (testicles of pigs.) Genotoxicity estimation in vitro for metal nanoparticles has been analyzed by the Comet assay method using eukaryotic cell culture ST. For the estimation of the mutagenicity of metal nanoparticles was performed using Allium cepa root tip meristems (“Allium test”). Results and discussions. The full set of experimental investigations of metal nanoparticles (СuNP, AgNP, AuNP, FeNP) showed low level of their cytotoxic effect. Obtained results demonstrate the absence of genotoxic influence of СuNP, AgNP, AuNP, FeNP both under the conditions in vitro and in the experiments that includes with using of metabolic activation system.СuNP, AgNP, AuNP and FeNP do not cause increased the frequencies of chromosomal aberrations in the apical meristem of Allium cepa compared with control samples, and therefore do not cause mutagenic’ action. ZnNP possess genotoxic, cytotoxic properties and were characterized by high values of the mutagenicity.Conclusions and prospects for further research. Investigated СuNP, AgNP, AuNP and FeNP are biosafe according to the genotoxicity, cytotoxicity and mutagenicity parameters. Fulfilled investigations open perspectives for using of investigated metal nanoparicles for modification of veterinary immunological means.
