Somatic embryo differentiation and germination of Coffea arabica L. cv. Catimor 9722 obtained in an orbital shaker

Abstract

The present work had as objectives to study the effect of four inoculation densities on the process of differentiation of embryogenic cell suspensions of Coffea arabica cv. Catimor 9722 and to evaluate the effect of two culture media described in the literature for the germination of somatic embryos. The results showed that somatic embryos were only formed in the treatments inoculated with 0.5 and 1.0 g FW l -1 , while in the treatments inoculated with 5.0 and 10.0 g FW l -1 only the cell aggregates were multiplied. The largest number of somatic embryos (23 154 SE.l -1 ) was obtained when inoculating 1.0 g FW l -1 , with statistical differences regarding the treatment inoculated with 0.5 g FW l -1 , in which 12 507 SE.l -1 were obtained. Regarding the germination of the somatic embryos in torpedo phase, the best results were possible when using 0.225 mg.l -1 de 6-BAP, because after eight weeks of culture they germinated with an average of 2.87 somatic embryos out of five placed initially in the culture flasks, what represented 57.5% of germination.
