Synthesis of Apocynin Dimer Derivatives Combining L-cysteine and α-lipoic Acid

Abstract

NADPH oxidase is a chief enzyme to regulate the production of reactive oxygen species (ROS), which has been regarded to be one of the important factors in development of some diseases, like stroke, asthma, acute lung injury, diabetes. Antioxidants are considered to be a group of potential therapeutics for treatment of these oxidative stress related diseases. In many researches, apocynin was reported to specifically inhibit NADPH oxidase activity, reduce ROS production and play a role against inflammation. In previous work, we synthesized JJA-D0, an analogue of apocynin dimer (diapocynin), which showed better NADPH oxidase inhibition activity than apocynin. Recently, hydrogen sulfide has been found to participate in various biological and pathological process, including anti-oxidative and anti-inflammatory injury. L-cysteine is an endogenous source of H2S production and α-lipoic acid is a well-known biogenic antioxidant. In this work, cysteine and α-lipoic acid were combined with JJA-D0, and six novel dimeric apocynin derivatives were yielded for sake of lead compound of better activity against inflammation and oxidation.
