The Investigation of Mycobacterium Bovis in Raw Milk Samples in Düzce Province

Abstract

Aim: Human tuberculosis caused by Mycobacterium bovis (M. bovis) is an important public health problem, especially in developing countries. M. bovis infections are often transmit from animal to human, rarely human to human. Contamination is achieved by raw consumption of contaminating milk and by the inhalation of contaminant aerosols. Tuberculosis cases caused by M. bovis were detected in a previous study conducted in our region. Therefore, it was aimed to investigate the presence of M. bovis in raw cattle milk samples in our study. Materials and Methods: In the study, raw milk sample collected randomly from 100 different cattle from five villages in Düzce province were examined, between 12-19 May 2017. Mycobacteria was investigated by Ehrlich-Ziehl-Neelsen staining method, culture on Löwenstein-Jensen agar, Middlebrook 7H11 agar and Mycobacteria Growth Indicator Tube medium, in these samples. In the case of presence of acid-resistant bacilli, it is planned to classify by conventional and molecular methods. Further, the maintenance conditions of cattle were observed. Results: When the incubation was complete, no acid-resistant bacilli were found in any of the colonies detected in 22 different milk samples, by Ehrlich-Ziehl-Neelsen and Gram stain. Gram positive bacilli, cocci and yeast were detected as contaminants. It was observed that the cattle in the ranch were in proper conditions. Conclusion: As a result, it was observed that the maintenance of the cattle was good in the villages in our province and cattle milks did not contain Mycobacterium. Because of the M. bovis cases previously identified in the region, it was thought that the study should be carried out in the fattening cattle as well.
