A preliminary study on genetic diversity of Asarum siebldii Miq.

Abstract

The genetic diversity of Asarum sieboldii Miq. is preliminarily analyzed with the approach of AFLP molecular marker. 1334 bands are accounted in nine populations by using 25 pairs of primer, 318 bands out of which are polymorphic. The percentage of polymorphic bands (PPB) is 23.84%. The mean Jaccard similarity coefficients and Nei genetic distances are 0.943 and 0.30 respectively. UPGMA analysis shows that the nine populations are gathered in two clades at 0.93 level of Jaccard similarity coefficient and 0.04 level of Nei genetic distance respectively, with six of them from eastern China in one clade and the rest from Qinling-Dabashan district in the other one. Z-test analysis based on the matrices of Jaccard similarity coefficients and Nei genetic distances show that the phenetic similarity relationships and the genetic relationships are of strongly coherent. The results indicate that the genetic diversity of A. sieboldii is very low and the populations have been differentiated to some extend. Moreover, the differentiation pattern of the populations is positively correlated with geographical distances. It is discussed that the population size of the species has sharply decreased because of human beings activity. The bottleneck effect, inbreeding depression and genetic drift resulting from population size decrease maybe the main reason for low genetic diversity of A. sieboldii.
