A selective fluorescent sensor based on Rhodamine<br /> for imaging mercury in living cells<br />

Abstract

Based on Rhodamine, a mercury fluorescent sensor R-A was described. The sensor contains two carboxyl-thiol metal-bonding moiety which exhibits highly selective response of fluorescence enhancement toward Hg2+ over other alkali and alkaline earth metal ions, transition and heavy metal ions in HEPES buffer solution. Furthermore, R-A has good coordination ability with Hg2+. Varies anions have no interference on the fluorescence enhancement with R-A, and R-A can detect micromolar level of Hg2+ in aqueous solution. The fluorescence enhancement of R-A towards Hg2+ maintains stable in wide pH span aqueous solution. R-E, the membrane-permeable ethyl ester of R-A, was successfully applied to tracking Hg2+ levels in Hela Cells and PC12 cells using confocal microscopy.
