小檗碱对Aβ25-35诱导大鼠PC12细胞<br /> 损伤的保护作用<br />

Abstract
目的：探索小檗碱对β-淀粉样蛋白25-35（Aβ25-35）诱导肾上腺嗜铬细胞瘤（PC12）细胞损伤的作用。方法：采用25 μM Aβ25-35处理PC12细胞36 h建立细胞损伤模型，应用细胞增殖抑制实验（MTT）评价细胞存活率，并检测细胞谷胱甘肽过氧化物酶（GSH-Px）和超氧化物歧化酶（SOD）的活性、细胞内活性氧（ROS）和胞内游离钙浓度（[Ca2+]i）的变化以及细胞凋亡。结果：给予Aβ25-35后，胞内GSH-Px和SOD活性降低，细胞内ROS含量和[Ca2+]i增加，并表现出凋亡的形态和特征，与凋亡相关的caspase-3蛋白表达明显升高。小檗碱（0.01 -10 μM）预处理24小时，再给予25 μM Aβ25-35后，细胞存活率增加、GSH-Px、SOD活性升高、细胞内ROS含量降低、[Ca2+]i降低、凋亡细胞数减少和caspase-3蛋白表达明显下降。结论：小檗碱对Aβ25-35诱导的PC12细胞损伤具有明显保护作用，其机理可能与其抗氧化、拮抗[Ca2+]i和抑制细胞凋亡有关。
