DESIGN AND IN-VITRO EVALUATION OF MUCOADHESIVE BUCCAL TABLETS OF REPAGLINIDE

Abstract

The goal of the present investigation was to design and evaluate mucoadhesive buccal tablet of Repaglinide for the treatment of diabetes (Type-II Diabetes mellitus). The tablets were prepared by using different polymers like Carbopol 934P, Hydroxy Propyl Methyl Cellulose (HPMC K4M), Sodium Carboxy Methyl Cellulose (NaCMC) and Hydroxy Ethyl Cellulose (HEC) as mucoadhesive polymer. Total nine formulations were developed with varying concentration of polymers. The carbopol 934P was used as primary polymer because of excellent mucoadhesive property and secondary polymer like HPMC K4M, NaCMC and HEC were used. The tablets were evaluated for hardness, thickness, weight variation, content uniformity, surface pH, swelling index, bio-adhesive strength, stability studies and in-vitro drug release. FTIR studies showed no evidence on interaction between drug and polymers. The surface pH, bio-adhesive strength and swelling index were found to be 6.5, 36.5gm and 99.53% respectively. Formulation F7 showed maximum drug release of 98.2% in 6 hour. The mechanism and order of drug release from the tablets were non-fickian diffusion and first order kinetics respectively. The results indicate that, the suitable mucoadhesive buccal tablets with desired properties could be prepared.
