CELLULAR INTERNALIZATION OF NANOPARTICLES

Abstract
Nanoparticles are becoming popular with drug delivery and drug targeting in cell biology because of their ability to overcome cellular barriers, ease to manufacture, stability, drug release pattern in controlled and sustained manner and ease of excretion of biodegradable polymers from biological systems. The therapeutic substances are encapsulated in surfactant and or lipid vesicles or in different polymeric system to prepare nanoparticles. These are able to attach with specific ligands and target to specific sites like mononuclear phagocytes, dendritic cells, endothelial cells, tumor cells and tumor neovasculature or cancerous cells in cellular environment. Cellular uptake and intracellular trafficking of nanoparticles occur in biological system through different mechanisms e.g. phagocytic and non phagocytic pathways and depend on their physicochemical characteristics. This study is reviewed on routes of administration of nanoparticles, different mechanisms of their cellular internalization particularly clathrin and caveola mediated endocytosis and their size, shape, surface property affecting the cellular uptake process.
