Detection of dsRNAs in Sclerotinia sclerotiorum isolates from Iran

Abstract

Sclerotinia sclerotiorumthe causal agent of sclerotinia stem rot is one of the most important fungal plant pathogens with global distribution. Broad host range and causing severe damage on crops annually, has been increased the incentive for further study of this species. Nowadays possibility of using mycoviruses as modern strategies of fungal biological control is important.From agriculturalperspectives, mycoviruses may contribute tosustainable agriculture as biological controlagents.The presence of viral dsRNA were investigated in 50 isolates of S. sclerotiorum. Isolates obtained fromdifferent hosts including lettuce, sunflower, cabbage, rape-seed, soybean, tobacco and Artemisia in north of Iran. The dsRNA molecules were separated using STE buffer, Phenol, Chloroform and column chromatography of cellulose (CF-11). Out of 50, 23 isolates were detectedwhich have dsRNA molecules.This is the first report of S. sclerotiorum dsRNAs from Iran.The impact ofmycoviruseson themorphologicalcharacteristics offungus was studied. ThedsRNA-containing had less mycelium linear growth thandsRNA-free isolates.
