DNP induced oxidative stress on blood components ameliorated by Pyrrole derivative of Tinospora cordifolia
Journal Title: Journal of Applied Biology & Biotechnology - Year 2017, Vol 5, Issue 1
Abstract
The intoxications and deaths caused by 2, 4-dinitrophenol (DNP) exposure through drugs, food supplements have been frequently reported and no specific treatment has been implemented to the toxic effects of DNP in adults and children. In this study, the purified compound (TCCP), a bioactive molecule from Tinospora cordifolia exhibited radical scavenging activities. Hence, we investigated the in vitro effect of TCCP on oxidative stress induced by DNP. Blood samples from healthy donors were treated with DNP (100µM/ml) for 2h. The DNP induced oxidative damage and effect of TCCP pre-treatment were determined by assessing the serum levels of cytoplasmic, lysosomal and oxidative stress markers. TCCP effectively inhibited DNP-induced oxidative stress on blood components, which was evidenced by the levels of endogenously generated reactive oxygen species and hydrogen peroxide. In addition, the depletion of GSH (reduced glutathione) content in DNP-treated cells was prevented by TCCP. Also, experimental results reflected the decreased expression of inflammation marker such as lipid peroxidase, catalase, LDH (lactate dehydrogenase), and phosphatase levels in cells with TCCP pre incubated DNP treated group. Therefore, these studies suggest the use of TCCP as an antioxidant and anti-inflammatory agent.
In vitro propagation by axillary shoot proliferation, assessment of antioxidant activity, and genetic fidelity of micropropagated Paederia foetida L.
An efficient plant regeneration protocol through axillary shoot proliferation of nodal explants of a valuable medicinal plant, Paederia foetida, has been developed. Highest shoot multiplication was found on Murashige and...
The Characterization of Amylolytic Enzyme Present in Fermented Sweet Sap of Palmyrah
Amylase enzyme obtained from 12h of naturally fermented sweet sap of palmyrah. The amylase showed zero order kinetics for 10min. The optimum temperature for the activities of enzyme as 65oC and pH was 7.0. When the enzym...
Human enterovirus A71 (EV-A71) isolated from acute flaccid paralysis patients without symptoms of hand foot and mouth disease
Background: Human enterovirus A-71 (EV-A71) is one of the causative agents for hand, foot, and mouth disease (HFMD). It is also reported to cause acute flaccid paralysis (AFP) during HFMD outbreaks and very rare to isola...
Isolation and Mycelial Growth of Mushrooms on Different Yam-based Culture Media
This study reports the potential application of yams as an efficient substrate for isolating mushrooms from the fruiting body. It seems that the utilization of yams as the medium for edible mushroom cultivation, i.e. a s...
Computational identification of miRNAs and their targets from Niger (Guizotia abyssinica)
MicroRNAs play a pivotal role in regulating a broad range of biological processes, acting by cleaving mRNAs or by translational repression. A group of plant microRNAs are evolutionarily conserved; however, others are exp...
