Effect of casting solvent on permeability of Anastrazole through Eudragit RL100 films

Abstract
In the present work, Eudragit RL100 films were prepared by using various casting solvents such as Acetone, chloroform, dichloromethane and ethyl acetate. The influence of solvent on the diffusion characteristics of drug through films were evaluated. To provide plasticity, n-Dibutyl phthalate at a concentration of 15% w/w of the polymer was used as a plasticizer in the preparation of films. The Solvent casting technique was used for the preparation of films. The prepared films were evaluated to check their suitability for use as a rate controlling membrane. The correlation coefficient values (r) revealed that the diffusion profiles followed zero order kinetics and the mechanism of drug diffusion was directed by peppas model. Based on water vapour transmission coefficient (Q) and Drug diffusion rate values the solvents can be ranked as in all the films; Ethylacetate- > acetone > dichloromethane > chloroform. The diffusion exponent of diffusion profiles (slope) has a value of n>1, which indicates super case-II transport diffusion. The results obtained in the present study revealed that the solvents used in the preparation of transdermal films have been shown considerable effect on the water vapour transmission and drug diffusion of the films. Eudragit RL100 films employed with ethyl acetate as casting solvent shown high diffusion rate when compared to other films.
