Endogenne mechanizmy powstawania reaktywnych form tlenu (ROS)

Abstract
Główne źródła reaktywnych form tlenu (ROS) w komórkach to: mitochondrialny łańcuch oddechowy i wybuch oddechowy fagocytów z udziałem oksydazy NADPH. ROS są wytwarzane również w retikulum endoplazmatycznym, peroksysomach, z udziałem oksydazy ksantynowej i oksydazy śródbłonkowej oraz w procesie autooksydacji małych molekuł.W warunkach prawidłowych ROS powstają jako produkt uboczny wieloetapowego procesu fosforylacji oksydacyjnej prowadzącej do syntezy ATP. Wytwarzanie ROS w czasie aktywowanego (np. zakażeniem) wybuchu oddechowego fagocytów odbywa się również wieloetapowo. Głównym enzymem odpowiedzialnym za to zjawisko jest oksydaza NADPH, a powstające ROS są wykorzystywane do walki organizmu z infekcjami.W opracowaniu omówiono aktualny stan wiedzy o budowie i funkcji mitochondrialnego łańcucha transportu elektronów, w którym produktem ubocznym są ROS oraz o budowie oksydazy NADPH, a także funkcji poszczególnych jej elementów w wybuchu oddechowym fagocytów.
