Fabrication, Characterisation and Evaluation of Rivastigmine Loaded Transdermal Films for the Patients Enduring with Alzheimer’s disease

Abstract

Alzheimer’s disease is the most common type of dementia which makes up 50 – 70% cases. Various cholinesterase inhibitors are used to treat conditions related to dementia. Among many drugs Rivastigmine is most promising in treatment of Alzheimer’s disease. In spite of therapeutic efficacy the drug has very short biological half-life due to which frequent dose administration is essential in order to avoid mood fluctuations. Besides its short biological half-life the drug has gastrointestinal adverse effects such as nausea and vomiting. In view with the above observed factors Rivastigmine loaded transdermal films were fabricated with 9.5 mg of dose per film per day. A total of 12 formulations with various polymers such as poly vinyl alcohol, ethyl cellulose, methylcellulose and HPMC were characterized and evaluated for thickness, flatness, % elongation, water vapor transmission studies, weight uniformity studies and drug release studies. Among the prepared formulations formulation F11 showed optimized parameters, the water vapor transmission test (WVT) values were observed between 1.30 – 1.70 mg cm-2.hr-1. Formulation F 11 has a WVT value of 1.32 mg cm-2.hr-1 .The % drug dissolved at the end of 24 hours was observed to be 95.8% for formulation F11 and it is further analyzed for drug release kinetics. From the kinetic studies the drug release is following zero order kinetics and the dissolution profile was best fit into Higuchi Model with regression value of 0.920. The formulation F11 is further analyzed for model independent kinetics such as similarity and difference factor and the results revealed that the test product is similar to that of marketed product.
