Study on the growth inhibitory effects of Puromycin and Doxorubicin on tumor cell lines (A549, HepG2, HT29 and K562) using Cell Titer-Glo and Alamar blue (Resazurin) based cell viability assays

Abstract

Cancer is a life-threatening spectrum of diseases in which mortality rates are high. There is more money being spent worldwide on In-vitro cancer research. Cell viability assay is the initial step to develop the anticancer drug. Here, we have used four different cancer cell lines (A549, HepG2, HT29 and K562) to understand the viability behaviour of different cancer against Puromycin and Doxorubicin. Before starting the vitality assay, cells were revived and maintained. Growth curve experiment were followed in 48 wells plate and counted for 4 days with every 24Hrs intervals. Haemocytometer was used for cell counting and based on the cell count data graph were plotted using prism software. CellTiter Glo viability assay and Resazurin viability assay were done to all the four cell lines. Envision luminescence plate reader used since both are luminescence based assays. Luminescence reading was converted to graphical data by using Prism software. Regarding the Growth curve doubling time is more to A549 and HT29 when compared to HepG2 and HT29. According to viability assays, IC50 of Puromycin and Doxorubicin were determined.
