Formulation and evaluation of fast dissolving Lamivudine oral thin films: an innovation in paediatric medication

Abstract

The present investigation under taken with objective of formulating Lamivudine fast dissolving oral thin films allowing fast reproducible drug dissolution in oral cavity thus bypassing the first pass metabolism to enhance the patient convenience and compliance. Lamivudine is used as antiretroviral drug and oral thin films of Lamivudine were prepared by solvent casting method with using different film forming agents like HPMC E15, HPMC 3cps, sodium alginate, xanthan gum, guar gum and their combinations. Propylene glycol, aspartame, orange flavour is used as a plasticizer, sweetening agent, flavouring agent and citric acid as saliva stimulating agent. OTF were evaluated for mechanical properties, weight variation, thickness, percent drug content, disintegration time, in vitro dissolution studies. Based on in vitro dissolution studies, Lamivudine with HPMC 3cps and sodium alginate combination (F6) showed the optimum performance over the other formulations which release the drug with in 45sec.
