Formulation and evaluation of floating bilayer tablets of propranolol hydrochloride

Abstract

Propranolol hydrochloride bi-layered, gastro-retentive floating tablets were formulated to increase the residence time of the drug by direct compression technique. The first layer of sustained layer was punched and second layer of floating layer was added and pressed by a single station press. Powders and finally pressed tablets were studied for micrometric properties, official evaluation tests for tablets were performed. Xanthan gum and sodium bicarbonate were the ingredients in floating layer and studied in 0.1mol/1ml HCl. The tablet rises to the surface and floats. The drug was slowly released without interference of gas bubbles. The swelling index, buoyancy lag-time and dissolution were studied. The release of the drug from the sustain layer is controlled by the amount of HPMC used. The floating capacity of the tablets was evaluated. The drug (propranolol hydrochloride) release from the tablets followed the Korsmeyer Peppas release model. The rate of drug release, buoyancy lag-time and swelling characteristics of the tablets was influenced by concentration of HPMC. The tablet was buoyant for 12hr. Formulation containing 80mg HPMCK4M seemed most desirable. The “n” value, 0.540 (0.5<n<1.0) of the formulation shows anomalous (non-Fickian) transport mechanism and is optimized. The FTIR analysis indicated the absence of any significant chemical interaction within drug and excipients. Stability study of optimal formulation proved that the formulation is stable.
