[i][/i] ISOLATION OF EXTRACELLULAR PROTEINS FROM SOIL

Abstract

 ABSTRACT The present research work was to investigate and isolate extracellular proteins three different farm soils. Soil samples were homogenized to remove large roots and stones, stored in dark at 4°C until use. The higher the pH of the phosphate buffer used for extraction, the larger the quantity of soil organic matter included in the prepared sample, resulting in a high background with unclear protein bands and tailing after SDS–PAGE. The results obtained suggested that some of soils derived from agricultural land contained significant amounts of a specific protein soluble in 67 mM phosphate buffer (pH 6.0). As a protein band is visible in electrophoretograms only when there is a sufficient amount of protein with the same apparent molecular mass. Key words: Extracellular proteins, SDS–PAGE, soil.
