In vitro fluconazole susceptibility of Candida parapsilosis strains isolated from various clinical materials

Abstract
Introduction: Candida parapsilosis (C. parapsilosis) is an important non-albicans species which infects hospitalized patients. The aim of the study: The aim of the present study was: assessment of susceptibility of Candida parapsilosis to fluconazole and evaluation of the influence of incubation time on the test result. Material and methods: 31 strains (one strain per patient) obtained from various clinical materials were studied. The strains were identified according to morphological and biochemical characteristics. E-tests were performed according to the manufacturer's instructions with a yeast suspension equivalent to 0.5 McFarland, Casitone agar plates (Bio-Rad) and E-test strips containing fluconazole 0.016 to 256 ug/ml (AB Biodisk). The plates were incubated at 35°C and read after24 and 48 h. The following breakpoints were applied: fluconazole susceptible - MIC64 ug/ml. QC was performed by testing C. parapsilosis ATCC 22019 and C. krusei ATCC 6258. Results: None of the C. parapsilosis strains in this study was found resistant to fluconazole after 24 h or 48 h of incubation. Two strains (6.45%) were susceptible-dose dependent but only after 48 h incubation. The fluconazole MICs for all strains were between 0.25-6.00 ug/ml after 24 h; and 1.00-32.00 ug/ml after 48 h of incubation. Conclusions: 1. No resistance was observed intested strains against fluconazole. 2. The fluconazole MICs read after 24 h were lower than those obtained after 48 h of incubation.
