Isolation, Identification and Drug Sensitivity Test of 3 Strains of Escherichia coli from Pet Dog

Abstract

In order to analyze the sensitivity of Escherichia coli ( E. coli) isolated from pet dogs to commonly used antimicrobial agents, anal swab samples from pet dogs in Luoyang pet hospital were collected to obtain three dog E. coli. The strain was isolated and cultured by Mac Conkey agar medium and eosin methylene blue agar selective medium. The E. coli DNA was extracted by boiling method, and 16S rDNA of E. coli was amplified by polymerase chain reaction ( PCR) method. The size of PCR products were detected by agarose gel electrophoresis and the amplification products were sequenced. The homology of E. coli was compared and analyzed by BLAST software, and the strain was identified as E. coli. The Kirby-Bauer method was used to carry out drug sensitivity test of selected 20 antimicrobial agents. The results show that E. coli isolated from pet dog can resist to 16 kinds of antibacterial drugs, such as ampicillin, gentamicin amoxicillin and ciprofloxacin. Wide drug resistance spectrum and multiple resistant bacteria are present.
