MRI-based quantification of grey matter reduction in brain areas along neural pathways in schizophrenia

Abstract
Schizophrenia is a critical disorder with a difficult diagnosis and no promising treatment. Therefore, it is important to address the neurological variation and the brain abnormalities linked to schizophrenia. In this paper, with the help of Structure MRI scans of 28 schizophrenia patients and an equal number of healthy subjects, a relationship between neurotransmitters, grey matter and antipsychotic drugs is established. Four major neurotransmitters are dopamine, glutamate, GABA and serotonin. To identify the changes in the grey matter, we used voxel-based morphology and statistical analysis in the experiment. As a result, we have found major grey matter reduction in schizophrenia patients. The brain areas impacted vary across several neurological pathways. The frontal lobe, limbic lobe, hypothalamus, caudate, putamen, substantia nigra, and lateral globus pallidus all showed severe shrinkage in dopamine. The midbrain, nucleus accumbens, brainstem, and frontal lobe all showed signs of serotonin-related atrophy. Glutamate and GABA-related areas, such as the cingulate gyrus, hippocampus, amygdala, brainstem, caudate, lentiform nucleus, putamen, superior frontal gyrus, thalamus, hypothalamus, occipital lobe, and parietal lobe, showed subtle grey matter changes. Our findings underscore the importance of neurotransmitter pathways and their links to grey matter shrinkage in schizophrenia. The observed brain areas provide light on the pathogenesis of the disorder and the influence of antipsychotic drugs on the brain. These results might help to create new therapeutic techniques that target specific neurotransmitter systems to prevent or delay grey matter degeneration.
