Optimal concentration of Ethyl Methane Sulfonate for phenotypic variation induction in Phaseolus vulgaris L. cv. ‘DOR 364’
Journal Title: Biotecnología Vegetal - Year 2016, Vol 16, Issue 3
Abstract
Common bean (Phaseolus vulgaris L.) is the main legume used for human consumption worldwide and is an important objective in breeding programs to favor certain characteristics of agronomic interest. In order to determine the optimal concentration of ethyl methane sulfonate to induce phenotypic variation in Phaseolus vulgaris L. cv. ‘DOR 364’ a study was carried out. The seeds were treated with different concentrations of ethyl methane sulfonate (0, 20, 30, 40, 50, 60 mM) and seeded in polyethylene bags containing 4 kg of substrate. Evaluations were carried out on variables involved in germination, growth, foliar development, plant morphology as well as yield components. The ethyl methane sulfonate influenced the different stages of development of the culture that were analyzed. Based on germination and plant development, survival, yield components and frequency of phenotypic variations, 30 mM ethyl methane sulfonate was selected as the optimal concentration to establish a population of possible mutants in common bean cv. ‘DOR 364’. To date, no previous studies have been reported. With the optimum concentration it was possible to generate the greatest number of phenotypic variations in the analyzed population.

Evaluation of the effect of Chitosan in the in vitro rooting stage of potato var. ‘Desirée’ and in the production of minitubers in greenhouse
The plantation of potato in vitro plants in greenhouse present some problems. Mainly the low uniformity of growing populations, the low and irregular yield and the low quality of the minitubers. The Chitosan is an elicit...
Identification and control of Bacillus sp., Contaminant of in vitro establishment of Guadua angustifolia Kunth
Efficient protocols of Guadua angustifolia Kunth micropropagation are not reported due to two principal problems for establishing it: explants necrosis and microbial contamination. In this work a high frequent bacterial...
Effect of adding different concentrations of activated carbon on the in vitro multiplication of yams
Phenolic oxidation can cause serious problems in the establishment, multiplication and survival of explants on most of plant species. The present investigation was developed toevaluate the effect of adding different conc...
Callus formation and somatic embryos of Cocos nucifera L. cv. Yellow Indian in semi-solid culture medium
The objective of this work was evaluated the effect of the concentration of 2,4-D and the type of explant in the formation of callus of Cocos nucifera L. cv. Indio Amarillo. The biggest percentages of explants with callu...
Comparison of three total RNA extraction methods from cacao leaves
The establishment of a method for extracting total RNA in plants that meets the required criteria of purity and yield is a critical step for subsequent molecular studies, especially in recalcitrant plants with naturally...
