Optimization of Shake Flask-ferment Conditions for Bacillus Lichenniformis B. LY02 Producing Cellulase

Abstract

In order to improve the production of cellulase from Bacillus lichenniformis LY02( B. LY02) strain,the shake flask-ferment conditions of cellulase produced by B. LY02 were optimized by using single factor experiments. The results show that the optimal carbon source is maltose and the optimal nitrogen source is yeast extract in B. LY02 fermentation medium. While the initial p H,cultivation temperature value,incubation time of B. LY02 fermentation medium is 5. 0,37 ℃ and 30 h respectively. Besides,loading medium volume is 60 m L( 250 m L triangular flask),initial inoculum is 2%,and the shaking speed is 180 r/min to 200 r/min. Under the optimal fermentation condition,the cellulase activity of B. LY02 was up to 0. 683 5 U / m L,which is increased by about 27. 73% than the original culture conditions.
