Rapid diagnosis of bacterial types of oral and maxillofacial space infection based on gas chromatography-mass spectrometry

Abstract

Objective To explore a rapid diagnostic method for bacterial types of oral and maxillofacial space infection (OMSI) based on gas chromatography-mass spectrometry (GC-MS) . Methods Staphylococcus aureus ( SA), a common bacteria of OMSI, was selected as the research object. The volatile organic compounds (VOCs) produced by the bacteria were detected by GC-MS. The bacterial volatile organic compounds (bVOCs) of SA were obtained after comparison with the background compounds in blank medium. The pus of 4 patients with OMSI were randomly selected clinically. After gradient centrifugation, the count and precipitation of bacteria were obtained. After detection, the patient volatile organic compounds ( pVOCs) were obtained. By comparing the peak areas of the same compounds, the probability of clinical infection with SA was calculated. Results The testing time after obtaining clinical samples was about 24 h. A total of 503 bVOCs were detected in SA standard substance, and the top 18 bVOCs were selected as the characteristic targets according to peak concentration and variable importance in projection. PVOCs and the 18 bVOCs were calculated to show that the probability of SA infection in 4 patients was 27% , 40% , 73% , and 40% , respectively. The actual results of clinical bacterial culture were as follows: undetected, Lactococcus lactis, SA, and Abiotrophia adjacens. Conclusion The diagnostic time of bacterial types can be greatly shortened by GC-MS technology. The diagnostic results of bacterial types based on GC-MS technology are consistent with the clinical bacterial culture results. Besides, GC-MS technology can also provide theoretical and practical basis for rapid diagnosis of bacterial types of maxillofacial infections during wartime.
