Tiroid Serbestleştirici Hormon Reseptöründe Nokta Mutasyon Eğilimi

Abstract
Giriş: Tiroid serbestleştirici hormon (TRH) enerji dengesini sağlamayı kontrolde önemli bir role sahiptir. TRH reseptörünün (TRHR) yapı ve işlevinde olabilecek patolojik değişiklikler genetik nöroendokrin defektin önemli bir nedenini oluşturur. TRHR içinde bir mutasyon görünebilir. Günümüzde, proteomik ve yapısal genomik çağ gereği protein nano-yapısı ve işlevinin artık tahmin edilebilmesi büyük bir ilerlemedir. Yöntem: Hastalıkların genomik ve proteomik düzeydeki bozukluklarıyla ilgili bilgiyi genişletmek üzere yeni bir yol olarak mutasyona açık noktaların belirlenmesidir. Burada yazar, TRHR amino asit sekansının peptid motiflerinin pozisyon eğilimlerini çalışmak üzere bir biyoinformatik analiz gerçekleştirmiştir. TRHR içindeki zayıf zinciri ortaya çıkartmak üzere GlobPlot adı verilen bir yeni biyoinformatik cihazı kullanıldı. Sonuçlar: Bu çalışmaya göre mutasyona duyarlı bölgeler saptandı. Yargı: Bunun anlamı TRHR'nin genetik olarak çok yüksek oranda sabit olmayan bir molekül olduğudur. Binlerce tip mutasyon beklenebilir. Klinikte yalnızca birkaç mutasyon eğilimi bildirilmiştir. Bu nedenle birçok gizli kalmış mutasyon eğiliminin saptanması olası olabilir.
